Argine has three pKa values.

i) Terminal carboxy group: COOH COO-, pKa1 = 2.17

ii) Terminal amine group: NH3+ NH2, pKa2 = 9.04

iii) Side chain group: NH2+ NH, pKa3 = 12.48

Net charge at pH = 9

pH > pKa1, so the equilibrium 1 shifts to right side, i.e. towards COO- (-1 charge).

pH < pKa2 and pH < pKa3, so both the equilibria 2 and 3 shift to left side, i.e. towards NH3+ and NH2+, respectively, (+1, +1 charges).

Therefore, the net charge for arginine at pH 9 = -1 + 1 + 1 = +1
