Are column chromatography and paper chromatography good procedures for separating, purifying and identifying compounds? Is sample size an important consideration?

Answer #1

Chromatography is a good technique to purify the compounds having the different polarity with separable Rf values.

Different polar compounds have an ability to show different Rf values.

Out of two Column chromatography is the best method to separate the large quantity of the compounds. Whereas, paper chromatography (thin layer chromatography) is used to identify the polarity system to the column chromatography or it can also be useful to separate coloured compound with small quantity.
