According to Micheal Menton equation for enzyme catalysis,
Rate of enzyme catalysis,R = K2 [E°][S°]/Km+[S°]
where K2 is turnover number
Km is micheal menton constant
[E°] is initial enzyme concentration
[S°] is intial substrate concentration
Now at high substrate concentration for chymotrypsin enzyme catalysis, [S°] >>>Km
So Km can be neglected in denominator, so that
Rate , R =K2 [E°][S°]/[S°]
R= K2 [E°]
This implies that rate is ZERO ORDER with respect to substrate concentration at high substrate concentration.
