No we cannot measure internal energy directly. Internal energy is the energy contained in the system and it is expressed im terms of change (delta). It depends on certain factors like kinetic anf potential energies which contribute to the total energy of the system. This can also be concluded from first law of thermodynamics,

where delta U is the change in internal energy, q is heat added on the system or by the system and W is work done on the system or by the system.
