What effect would changing to HILIC have on elution order of the parabens? What effect does heating the column have on resolution for HPLC?
HLIC mode utilises the difference of hydrophilic properties between the compounds. The order followed for elution os least polar to more polar. Parabens are esters of para-hydroxybenzoic. Polarity decreases when the hydrocarbon chain increases on R. Increase in hydrocarbon chain will decrease the polarity as hydrocarbon chain forces between water and molecule and breaks the hydrogen bonding making them less polar. So parabens with long hydrocarbon chain will elute first then ones with a shorter chain.
On heating, the column the solute transfer from mobile phase to stationary phase is more efficient which gives high efficiency even at elevated flow. the low pressure will also help to increase resolution and efficiency. Heating the column will prevent temperature gradient occurrence GIVING IT HIGHER RESOLUTION.
