A) We can measure the disappearance of Z at 340 nm (absorbance will decrease), or we can measure the increase in Y at 480 nm (absorbance will increase). This will not measure the enzyme, but will measure the enzyme activity.
B) Turnover number = Vmax/Et

Vmax is the maximum velocity and Et is total amount of enzyme
Vmax = 60 µ mol/min

Et = 0.4mL x 0.1mg/mL = 0.04 mg enzyme    (400 µL = 0.4 mL)
Turnover number = (60 µmol/min) /(0.04 mg)
Turnover number =1500 µmol/min/mg
