A certain protein can bind one of two ligands: ligand A or ligand B. below is a plot of the fractional saturation (Y) versus ligand for ligand A and ligand B. How do I calculate approximate Kd? for example if the ligand is 200 uM and the fractional saturation is 65%?

Answer #1

Given, [L] = 200 M = 200 x 10-6 M   , fractional saturation = 65 % = 65 /100 = 0.65

We have the relation as--

fractional saturation = [L] / ([L] + Kd)

=> 0.65 = 200 x 10-6 / ( 200 x 10-6 + Kd)

=> 0.65 * ( 200 x 10-6 + Kd) = 200 x 10-6

=> 130 x 10-6 + 0.65 Kd = 200 x 10-6

=> 0.65 Kd =( 200 x 10-6 ) - (130 x 10-6)

=> 0.65 Kd = 10-6(200 - 130)

=> 0.65 Kd = 70 x 10-6

=> Kd = 70 x 10-6 / 0.65

=> Kd = 107.69 x 10-6

=> Kd = 1.07 x 10-4

17. Which of the following statements are accurate? A. Neurotransmitters can act as ligands. B. Acetylcholine is a neurotransmitter. It can bind to an acetylcholine receptor on the surface of a cell. If this receptor is also a sodium channel, we would call acetylcholine a ligand and its receptor a ligand-gated receptor. C. Neurotransmitters can act as chemical signals between two neurons. D. Diffusion is a type of passive transport and does not require energy. Sodium channels are an example...

An ion-channel is a transmembrane protein that can be in one of two states: OPEN (which allows ions to pass through the membrane) and CLOSED (which does not permit ions to pass through). You have an ion-channel in which the energy of the open state is 600 cal mol-1, the energy of the closed state is 1800 cal mol-1, and the temperature of the system is 300 K. Below are ten functions. Find the first derivative of each. All of the derivatives can be found by using combinations of the constant rule, power function rule and sum-difference rule.   Do not use the product rule. It is not needed. The degree of difficulty (more or less) increases from (a) to (j). Be sure to show intermediate work. Five points are awarded for correctly developed derivatives. There is no partial credit, because an incorrect derivative is useless. For example,...
