A common biological buffer is a "Phosphate Buffer," which involves the equilibrium between H2PO4- and H2PO42- , with a pKa at 25 degree celcius of 6.86. Describe in words how you would prepare 500.mL of 0.250 M phosphate buffer at pH= 7.2, given appropiate glassware and 500.mL each of 0.250 M NaH2PO4 and 0.250 M Na2HPO4 at 250 C. (Note: your explanation needs to include actual volumes, so this question requires calculations! It will be helpful to know that the concentration of each component must add to the buffer concentration. i.e. [A-] + [HA]= 0.250 M).

Please use both words and calculation to understand.

3.Phosphate-buffered saline (PBS) is a buffer solution commonly used in biological research. Most formulations contain a mixture of KCl and NaCl buffered to a pH of 7.40 using phosphate-containing compounds. a.The following reagents are available in your lab. Choose the correct reagents and calculate the volumes necessary to prepare exactly 5.0 L of a phosphate buffer with a pH of 7.40.                                                0.100 M H3PO4 0.150 M Na2HPO4                                                0.200 M KH2PO4 0.120 M Na3PO4 b.Commercial PBS solutions are typically prepared...

Show calculation for the assigned buffer. Recall: [acid] + [base] = 0.20 M. Calculate the individual concentrations needed (mass or volume) for HA and A- based on your assigned buffer and pH. The condition [acid] + [base] = 0.20 M must be satisfied. The final volume of solution is 100.0 mL. Assigned Buffer: sodium dihydrogen phosphate and disodium hydrogen phosphate pH = 7.69 Supplemental Data (I believe Ka is 6.2 x10^-8) Acid Species Ka Molar Mass g/mol Compound Phosphoric density...
