Use of affinity column:
If the net charge on an isozyme is positive/negative, then the isozyme is acidic/basic, which if you are passing through affinity column, it will stick to the column due to the corresponding affinity, then its elution will be delayed.

Use of ion-exchange column:

If the net charge on an isozyme is positive/negative, then the isozyme is acidic/basic, which if you are passing through an cation/anion-exchange column, it will be exchange by cation/anion, then its elutions will be fast.
