You created a standard curve using chlorophyll a from spirulina rather than leaf extract since it doesn’t contain chlorophyll b, yet were later told chlorophyll b doesn’t fluoresce. What might have happened if you made a standard curve using crudely-purified chlorophyll a from leaf extract? How would it have affected your results and why?
The results of this experiment is dependent upon the concentration of chlorophyll a taken for the experiment. Since in leaf extract we would have possible other components along with chlorophyll a, the concentration assumed would be incorrect and low value of chlorophyll would actually be present, which would give low value than expected value for a particular run. This would lead to a error in meaurement and would not be reliable and can be considered as standard.
