Usually The polyhistidine tagged proteins bind to resins such as Ni-NTA (NTA=nitrotriacetate) which is the most commonly used ligand for IMAC (Immobilized metal-affinity chromatography is also known as metal-chelate affinity chromatography (MCAC). .) .  because this particular metal and chelation has better binding properties and suitability for specific applications of IMAC.

Histidine-tag purification uses the purification technique of immobilized metal affinity chromatography IMAC.Imidazole competes with the his-tag for binding to the metal-charged resin and thus is used for elution of the protein from an IMAC column.
