Draw each of the basic amino acids at ph 3.0, as in stomach acid. How may this change the way they behave in a folded protein?

Homework Answers

Answer #1

Histidine is the basic amino acid in stomach. The structure is

  • Amino acids interact with each other to produce a well-defined three-dimensional structure, the folded protein (the right hand side of the figure), known as the native state. The resulting three-dimensional structure is determined by the amino acid sequence
  • The essential fact of folding, however, remains that the amino acid sequence of each protein contains the information that specifies both the native structure and the pathway to attain that state. This is not to say that nearly identical amino acid sequences always fold similarly.
  • Histidine is also in the stomach. It is helpful in producing gastric juices, and people with a shortage of gastric juices or suffering from indigestion, may also benefit from this nutrient.

The structures of the amino acids leucine and isoleucine are shown: If a mutation in the DNA of a globular enzyme changed all of the leucine amino acids to isoleucine, predict how the relative position of the amino acid in the tertiary structure of the protein would be affected when the protein is placed in an aqueous solution. If the leucine amino acids were found within the active site of the enzyme, discuss the effect this change is likely to...

A 500 amino acid protein consists of 200 hydrophobic residues. Upon folding, 150 of the hydrophobic amino acids fold and form a hydrophobic core. Determine the ∆G o for the protein at 300K, considering contributions from both the protein and the water. Assume the following: When the protein folds, each amino acid (hydrophobic and hydrophilic) forms an interaction that is 3 kJ/mol more stable thant the unfolded state. In the unfolded state, the hydrophobic amino acids contact and influence the...
