Describe the preparation of 5 liter 0.3M acetate buffer pH 4.47 from 2M solution of acetic acid and 2.5M solution of KOH.
Ans. Add 750 mL of acetic acid stock solution + 200 mL KOH stock solution and bring to 5L with water
The molar ratio of the buffer components is calculated:
n Salt / n Acid = 10 ^ (pH - pKa) = 10 ^ (4.47 - 4.75) = 0.5
It has:
i) n Salt - 0.5 * n Acid = 0
ii) n Salt + n Acid = M * V = 0.3 M * 5 L = 1.5 mol
System of equations is applied:
n Salt = 0.5 mol
n Acid = 1 mol
The volume of each reagent is calculated:
V acid = n acid * 1000 / M = 1 * 1000/2 M = 500 mL
V KOH = 0.5 * 1000 / 2.5 M = 200 mL
