*Calculate how many grams you need to make a 20mM buffer of Tris/ 1mM EDTA buffer?...

*Calculate how many grams you need to make a 20mM buffer of Tris/ 1mM EDTA buffer? (1360 mL final volume) Tris:121.14 g/mol. EDTA:292.24 g/mol

Homework Answers

Answer #1

Molarity = Number of moles of Solute/Volume of Solution in L

20mM = Number of moles of solute/1.360L

Moles of Tris required = 27.2 m moles

Mass of Tris required = number of moles * molar mass = 27.2 m moles * 121.14 gm/mol = 3.30208 gms

1mM = Number of moles of solute/1.360L

Moles of EDTA required = 1.36 m moles

Mass of EDTA required = number of moles * molar mass = 1.36 m moles * 292.24 gm/mol = 0.39744 grams

Total mass of solute required = 3.30208 gms + 0.39744 gms = 3.699 grams

TAE is a commonly used buffer for DNA el ectrophoresis. This buffer contains 40 mM Tris, 20 mM Acetate, 1 mM EDTA as the final concentrations. A 50X stock solution (50X the final concentration) is normally prepared. You have Tris powder (mol. wt. 121.14), glacial acetate acid (a liquid with a density of 1.05 g/mL and a mol. wt. of 60) and 0.5 M EDTA stock solution. You need to make up 1 Li ter of 50X TAE.
