Bradford protein assay

It is based on the chemical reaction between Coomassie brilliant blue G-250 dye and proteins in solution

The dye binds to arginine, tryptophan, tyrosine, histidine, and phenylalanine residues and is highly sensitive

Direct protein assay

Direct protein assay needs no chemical reaction to be performed

This is based on the fact that the aromatic amino acids (tryptophan and tyrosine) show a peak in the UV-vis spectrum around 280 nm and has a moderate sensitivity only
