why must use semi log to graph protein migration in electrophoresis

Answer #1

The proteins will appear as blue bands. The size of the most abundant proteins can
be determined by setting up a standard curve using the size of the molecular weight
markers and the distance each marker band migrated from the well. Measure the distance
that each band of protien migrated from the bottom of the well to the middle
of each band . Measure the distance that the dye front migrated in each lane and record
CALCULATE THE RELATIVE MOBILITY (Rf) for each protein by dividing the distance migrated
by the distance the dye front migrated.the distaance each protien migrated the molecular
weight of each protien. Use semi log graph paper to graph the standard curve by placing the
molecular weight of each marker on the axis of the graph and Rf value on the X-AXIES

Imagine that you have just run protein gel electrophoresis using the same protein ladder that we used in this experiment as your molecular weight standard. In the test sample (in the lane just to the right of the ladder), after running the gel you stain for a protein band that is positioned almost exactly halfway between the Phosphorylase and BSA bands on the ladder. A colleague in the lab points to the band and says it must have a molecular...

The tertiary structure of a protein must also be considered when comparing deamidation rates. Another source of catalytic groups are amino acid side chains such as the ones discussed above that are located in the same vicinity as the labile Asn or Gln. Another consideration is the location of the Asn and Gln with respect to the tertiary structure of the protein. It has been observed that Asn and Gln residues in the interior of a protein are deamidated at...
