TRIS isba buffer used commonly with biological samples. what is the pH of a TRIS buffer when 0.23M acid is mixed in equal amounts with 0.67 M of TRIS base?

Homework Answers

Answer #1

For making of TRIS isba buffer,

TRIS BUFFER MATERIALS

  • TRIS powder
  • Hydrochloric acid
  • Distilled water
  • pH meter

For calculation of pH of buffer, acid is HCl and base is TRIS powder whose diagram is given below

The formula for finding pH of the buffer is :

pH = pKa + log [A-]/[HA]

for finding pKa, we have to know Ka of HCl = 1.3 * 106

  So, pKa = -log(1.3 * 106)= +6.1139

so, pH = +6.113 + log(0.67/0.23)

=-6.113 + log(2.913) =-6.113 + 0.46 =-5.653

Here we taken HCl as base.
