there is only one disulfide bond present. After treatment with trypsin, two polypeptide fragments were produced. After treatment with chymotrypsin, only one polypeptide was produced. in both treatments the disulfide bond remained intact.

The trypsin digestion site is C-terminal side of the positively charged amino acids, such as Lys and Arg, but no Pro after Lys/Arg.

The chymotrypsin digestion site is C-terminal side of the bulky hydrophobic amino acids, such as Phe, Trp, Tyr, but not Pro after Phe/Trp/Tyr.

Since there are 2 fragments are formed on trypsin digestion, no disulfide bond is possible between CYS1-CYS2, CYS1-CYS3, CYS1-CYS4

Since there is only a single fragment on chymotrypsin digestion, there should be disulfide bond between CYS2-CYS4 or CYS3-CYS4. No disulfide bond between CYS2-CYS3.
