Enzyme Kinetics: Plot the data below and calculate the kinetic parameters, Km and Vmax. You must submit the graph with your calculated parameters
[S] (uM) 0.1, 0.2, 0.4, 0.8, 1.6
vo (mM/s) 0.34, 0.53, 0.74, 0.91, 1.04
Km = 0.2543 uM
Vmax = 1.2048 mM/s
------------------------------
To determine the values of Km and Vmax, by using Michaelis and Menten Equation, Lineweaver-Burk plot is used in which 1/Vo vs 1/[S] are plotted.
The linear plot of the type y = mx + c can be related to the equation,
1/Vo = (Km/Vmax)(1/S) + 1/Vmax
m = Km/Vmax)
c = 1/Vmax
1/Vo Vs 1/S plot, give a linear fit with y = 0.2111x + 0.83
So,
Km/Vmax = 0.2111
1/Vmax = 0.83
Vmax = 1/0.83 = 1.2048
Km = Vmax x 0.2111 = 1.2048 x 0.2111 = 0.2543
