1. What changes to the migration pattern would you expect if the solvent system contained more water? More chloroform? TLC Analysis of Egg Lipid

Homework Answers

Answer #1

Egg lipid generally consists of three major components: lipids (less polar fatty compounds); cholesterol ( the most polar component due to OH groups); & palmitate (more polar than lipids)

T LC is a technique based on adsorption & desorption phenomenon. Less or non polar components are soluble in less or non polar solvents and dragged towards the solvent front. & polar components by polar solvents.

If water is more in mobile phase, the solvent becomes more polar& hence the polar components are eluted first.

If chloroform is more in amount, it dissolves non polar components in it & hence drags non polar lipids towards the solventfront.

Which solvent system is more polar 30% Acetone/70% Hexane OR Ethanol? How would you be able to tell this from a column chromatography? I am just confused about this. I think that it would be Ethanol. But I am unsure how to tell that from the column chromatography because wouldn't the more polar solvent take the beta carotene with it instead of the chlorophyll?

Ethyl acetate is another common organic solvent used in the type of TLC experiment you performed: it has a higher molecular weight and boiling point than either petroleum ether or diethyl ether, and is more polar than them as well.
