Typical kinetic studies are done with fixed amount of enzyme in a fixed volume with varied levels of substrate to obtain a value for Km and Vm. Does doubling the amount of enzyme in an assay change: Km? Vm? kcat? explain

Homework Answers

Answer #1

When the enzyme is saturated with substrate Vmax is the maximum rate of reaction .

As it depends on the enzyme concentration, if the amount of enzyme is doubled, Vmax is doubled. Changing the enzyme concentration will not change their value , Km and k cat are constants .

kcat is a constant. In enzyme kinetics, it describes the turnover rate of an enzyme-substrate complex to a product.

KM is the Michaelis constant. It gives the amount of substrate required by an enzyme to attain half of its maximum reaction rate.
