Question

Protocol to make 1% SDS

Protocol to make 1% SDS

Homework Answers

Answer #1

SDS means standard solution.

1%SDS means 1g of solute disolves in 100 ml of solvent.

Protocol :

1. Before going to weigh the sample, we should ensure whether the weighing machine is calibrated or not.

2. If it is calibrated, then go for weighing of the sample.

3. Ensure that whether the weighing machine is in air free zone or not.

4. Weigh 1g of the sample and transfer into a clean and dried standard flask.

5.Dissolve the contents by adding 50 ml of the solvent.

6.Stirr and Shake well until the contents completely dissolves in the solvent.

7.Now make up the solution up to the mark of the Standard flask.

8.Now the standard flask contains 1%SDS.

In a research lab, you will not always be given a protocol with the solutions you will need explicitly described. Often researchers have to adapt to the information that is provided in a published paper in order to make a solution. For the following questions assume you have been given the following information: 4X TGB (tris-glycine buffer) pH 8.3 Tris: 25mM Glycine: 250mM SDS: 0.1% Part A: How many milligrams of SDS will you add when making 500mL of 4X...

Sodium dodecylsulfate (SDS) is an ionic detergent. At high concentrations, it completely denatures a protein whether it is a soluble or membrane protein. It is for this reason that SDS is routinely used in SDS-polyacrylamide gel electrophoresis (SDS-PAGE) to allow the molecular weight of a protein to be accurately determined. Explain why SDS is a good detergent that can completely solubilize membrane proteins
