What type of substances are more suitably separated by paper chromatography than by thin-layer chromatography?
In paper chromatography the separation depends upon partition of substance between two phases and the adsorption effects of inert support on compounds undergoing chromatographic separation.In TLC the seperation depends on the Rf (Retardation factor) values. Generally separation of pigments present in the leaves (spinach) and flowers (rose, marigold) are done by paper chromatography than TLC (thin-layer chromatography). But TLC methods are using in these days for better performance. TLC can be dipped into ninhydrin or beta-napthol solutions and then charred to observe color for colorless compounds, whereas in paper chromatography this is not possible. Hence mostly colored compounds are used in paper chromatography, but iodine is sometimes used to identify.
