I wanted to reduce the levels of protein A in cells. Which small ncRNAs would I use?
We can use micro RNA or miRNA and sn RNA. These small non coding RNA's should have complementary sequence with respect to the target mRNA. Here the target RNA is our RNA which makes protein. These small non coding RNA binds to the target mRNA and converted into ds RNA. This will inhibit the ribosome to bind to target mRNA. Or this will leads to digestion of target mRNA by the Argonaute and RISC complex. Hence the protein synthesis of target mRNA is reduced.
