Explanation

During processing , kelp blade or bladder was washed, and then chopped up into a small amount of water. Later it was disrupted with the addition of a mixture of detergent and acetone. Then mixture was filtered and filtrate is taken to meaure in spectophotometer.

So the solution is having pigment in detergent and in acetone liquid. So we will use these two as blank. Because these two can give some absorbance value so that will be counted along with pigment absorbance value and will give false results. So first we will use these two as blank and after adjusting blank with these two , whatever absorbance sample will give , just due to the pigment.
