What would happen to TRANSLATION if there was a nonsense mutation in the 5’ untranslated region of an mRNA? Explain your reasoning

Homework Answers

Answer #1

There would be no effect on the translation process when the non sense mutation is found in 5' end of the untranslated region. The untranslated region provides the stability to mRNA only. This region is not translated into protein. The protein synthesis occurs between the start codon and stop codon. Hence if the mutation is found in between AUG and stop codon, then there is found effect of mutation only. That's why there is found normal translation of protein occurs.

You hypothesize that a miRNA you discovered regulates the translation (inhibits the translation) of an mRNA in adult C. Elegans. Briefly explain an experiment that you would perform to test your hypothesis that the miRNA has an effect on the translation of the mRNA in adult C. Elegans. You have isolated a nonsense suppressor that maps to a the tRNA-leu gene. If the suppressor has a single point mutation in it, and it suppresses the 5’ UAG stop codon then, (a) what is the sequence of the anticodon in the mutated tRNA-leu gene and what was the original sequence before mutation? Show your work (codon-anticodon base pairing for example). (b) Would this suppressor also suppress termination of the UAA stop codon? Explain.
