Chromatin immunoprecipitation assay is a technique for investigation whether specific proteins are associated with specific genomic regions. It is actually done to determine if any interactions are present between DNA and protein in a cell.
Steps are:-
Crosslinking of DNA to histone protein in the living cell and cell lysis to release chromatin.
⬇️
Fragmentation of chromatin with nuclease digestion and sonication.
⬇️
Enrichment of target chromatin using an antibody specific protein (Antibody- magnetic bead complex)
⬇️
Purification of associated DNA by removal of the protein with protease and elution of DNA.
⬇️
Analysis of DNA by polymerase chain reaction (PCR).
