A researcher needs to make 1X citrate buffer and finds a 7X solution of the buffer in the lab stock cabinet. How many mL dH2O should the researcher use to make 750 mL of the 1X buffer for the experiment? The buffer can be diluted with dH2O.

The working buffer should have 1 X concentration.

The final volume of working soution is 750 ml.

The volume of stock required is (V) :

7X × V = 1X × 750 ml

V = (1x × 750) /7X

V= 107.1428 ml

V= 107.14 ml

The stock buffer is taken of 107.14ml

Volume of distilled water used = 750- 107.14= 642.86 ml.

Hence 107.14 ml buffer is dissolved in 642.86 ml of distilled water in such a way that finam solution becomes 750 ml. In this way, the working solution of 1X of 750 ml is obtained.
