Microbiology Unknown Bacteria Sample: you received a mixed broth culture and were instructed to identify all organisms present in that mixture. What are the first steps you should do to accomplish your goal?

If u get an unkown mixed broth of culture, the first few steps done are as follows:-

1. Isolation- it is the first step done to isolate pure bacterial cultures. It can be done by various methods like streaking,dilution, plating etc.

2. Staining reaction-It is the basic technique used to identify microorganisms by using Methylene blue.

Gram staining os a differential staining technique to differentiate the bacterium whether it is gram positive or gram negetive.

3. Biochemical reactions: they are used to differentiate the members of enterobactericae. They are done by IMViC tests like indole test, methyl red test,Voges proskauer test and citrate test.
