Generally, the spread plate methods is used to inoculate the bacteria onto the agar plates. Disk diffusion method is probably the most widely used method for determining antimicrobial susceptibility testing. An antibiotic-impregnated disk, placed on agar previously inoculated with the test bacterium. The antibiotic diffuse radially outward through the agar medium producing an antibiotic concentration gradient. Zone of bacterial inhibition indicated the susceptibility of the bacteria for the antibiotic/antimicrobial agent.
