Material and methods used to grow tetrahymena vorax

Materials:

• Tetrahymena vorax strain( predator)
• Tetrahymena pyriformis strain (prey)
• Loefer's medium (for growth of T. vorax)
• Distilled water.
• Large petridishes.
• pH meter.

Method:

1. The populations of both T. pyriformis and T.vorax were grown on Loefer's medium for 48 hours.
2. They were then washed in distilled water.
3. The density of predator population was adjusted to one hundredth time of the density of prey population.
4. Equal volumes of prey and predator suspensions were mixed together.
5. For larger yields of macrostomes of T. vorax, large surface area is preferred such as petridishes.
6. pH is to be adjusted to 6.0; temperature to be set to around 20⁰C for around 12 hours.
7. Macrostomes of T. vorax strain are seen to appear after 6 hours of the addition of T. pyriformis strain.
