Primase

A short strech of 5-7 nucleotides de novo RNA primer or DNA primer is synthesized by primase enzyme. RNA primer is complementary to the template strand of DNA. DNA polymerase directly can't synthesize new strand but add nucleotides to the 3'end of the primer. Helicase enzyme opens the double stranded DNA and primer will bind with the complementary strand and DNA polymerase add nucleotides to the 3' end of the primer to synthesize new strand of DNA in 5'-3' direction. After complete synthesis of DNA strand RNA primer is replaced with the DNA.
