1. By increasing the amount of agarose in the gel, the distance travelled by the pieces of DNA will be reduced. Increasing the agarose concentration will lead to creation of smaller pores in the gel matrix. Due to this, the DNA will be able to migrate a shorter distance.
2. It is difficult to load the sample into the well because we are trying to place the sample into a clear well that is surrounded by a clear gel. Therefore the sample gets loaded into the gel instead of the well.
