A microbiologist performs oxidase test to check whether the organism produces cytochrome c oxidase or not.
Cytochrome c reduces the reagent Tetramethyl-p-phenylenediamine dihydrochloride to the purple coloured end product ( indophenols ).
Take a filter paper and add few drops of oxidase reagent on it. Now take the test organism and make a smear on the filter/blotting paper. Observe the colour change. If purple colour is produced then the organism is oxidase positive.
This test helps to distinguish between aerobic and anaerobic bacteria as only the aerobic bacteria have the ability to use oxygen as a terminal electron acceptor in respiration.
Oxidase positive bacteria : Pseudomonas, Vibrio, Neisseria, Brucella and Pasteurella .
Oxidase negative bacteria : Enterobacteriaceae are oxidase negative .
