A 10kb DNA molecule disgested with restriction endonuclease EcoRI yielded fragments 4 kb and 6 kb in size. When digested with BamHI, fragments 1, 3.5, and 5.5 kb were generated. Simultanous digestion with EcoRI and BamHI yielded fragments 0.5, 1, 3, and 5.5 kb in size.

Is it possible to determine whether the original molecule was linear or cicular? if not, why? if it is possible, why is it one or the other?

Answer #1

yes it can be known. you need to calculate both scenario,

IF it was circular, first digestion would mean there are 2 sites and second mean there are 3 sites making a total of 5 sites. so double digestion would result in 5 fragments but it is not 5 in double digestion.

If it was linear, the single digestion would mean there is only one site in first case and 2 sites in second case making a total of 3 sites of both ecor1 and bam h1 and both of them when used simultaneously should give 4 fragments which it is giving.

HENCE, the molecule is LINEAR.

During the trial, lawyers for the accused said that the men believed that the accounting decisions they made were appropriate at the time, and that the accounting treatment was approved by Nortel’s auditors from Deloitte & Touche. Judge Marrocco accepted these arguments. Marrocco added he was “not satisfied beyond a reasonable doubt” that the trio (i.e., Dunn, Beatty, and Gollogly) had “deliberately misrepresented” financial results. Given the facts of the case, do you believe Judge Marrocco’s decision was justified? Explain....
