A competitive inhibitor can be overcome by increasing the substrate concentration. The excess amount of substrate can negate the competitive inhibitor and the maximum velocity is ultimately unaffected. On the other hand, noncompetitive inhibition cannot be overcome by increasing the substrate concentration, and it depends on the concentration of inhibitors. Noncompetitive inhibitor may bind either to a free enzyme or to an enzyme–substrate complex, and this inhibition is relatively rare. Another compound that block the binding of noncompetitive inhibitor to the enzyme must be added to reduce the effect of the inhibitor.
