For enzymes & any other class of proteins: Given information regarding the amino acids in a binding site, create hypotheses regarding how binding of small molecules to the site can lead to changes in protein function and how factors such as substitutions of different amino acids within the binding site, pH and temperature would impact your hypotheses.

The active site (binding site) of the protein where the ligands form a temporary bonds with the binding site and brings conformational changes of a protein usually alteration in shape of the tertiary structure of a protein. This
conformational change brings the function of a protein.

Each protein has a specific amino acid sequence in the active site for their function. If different amino acids are substitue in a binding site, the protein totally lost its activity. Ligands are not able to form a bonds with active site.

pH and Temperature both impact in the alteration of tertiary structure of a protein.But it not brings any function
of a protein. It only leads to denature or instabilty or non function of a protein.
