How would you prepare 50mL of a 3% agarose with 1XTBE WHEN YOU ONLY HAVE A BOTTLE OF 10XTBE IN YOUR LAB? (Hint: You need to first dilute with dH2O your the 10XTBE to 1XTBE)

Answer #1

(ANSWER)

(EXPLANATION)

AIM - Prepare 50mL of a 3% agarose with 1XTBE.

First prepare 1X TBE.

STOCK CONCENTRATION is 10X TBE

FINAL CONCENTRATION is 1X TBE

FINAL VOLUME is 50ml

C1V1 = C2 V2

C1 = CONCENTRATION OF SOLUTION 1

V1 = VOLUME OF SOLUTION 1

C2= CONCENTRATION OF SOLUTION 2

V2 = VOLUME OF SOLUTION 2

10 x a = 1 x 50 ml

a = 50/10 = 5ml

5ml of 10X TBE is required to prepare 50ml of 1X TBE.

Preparation of 50ml of a 3% agarose in 1X TBE

3 % AGAROSE means 3grams of agarose is required for 100ml 1X TBE solution.

For 50ml of 1X TBE (3/100)*50 = 1.5 g

1.5g of agarose is required to prepare 3% agarose in 1X TBE.

Lab 9 – Molecular Biology In this lab, you will prepare an agarose gel and use gel electrophoresis to compare the size of 2 dye molecules Methyl orange and Ponceau G. You will also analyze an “unknown” sample that contains a mixture of two dyes. Dye molecules with lower molecular weight or greater electrical charge will migrate faster through the gel, than dye molecules with greater molecular weights or lesser electrical charge.
