Answer #1

Gel filtration chramatography is a technique which separates the proteein on the basis of their size. It is also known as gel permeation chromatography.

In this technique, stationary phase is made up of porous matrix and mobile phase is the buffer which flows between the matrix and beads. Beads have a defined range of pore size. So once protein get entered into the beads, it will restrict the movement of proteins. Higher the size of the protein faster would be the elution and vice versa because small protein get enetered into the beads and face more retention in the column.

So each and indiviual protein of different size will give a separate fraction, whihc can be collected manually.

We did a lab on Gel Filtration Chromatography in which we used two "standards" Phenol Red (included in gel) and Blue Dextran(excluded from gel). An unknown protein (either hemoglobin or myoglobin) was given. Question is why is a standard used (in relation to the lab) and how is it useful? Please provide a detailed answer. Thank You.

Experiment 2: Restriction Enzymes (Pop-It Bead Lab) Post-Lab Questions How can DNA be fragmented into very specific sections? Where do restriction enzymes come from? What is their function in nature? How do molecules of varying sizes separate in electrophoresis? What is the purpose of the gel? What about the electricity? Investigate one way in which electrophoresis is used in medicine today. Write one or two paragraphs to explain your findings.

How would I seperate benzophenone from benzoic acid in a under-grad lab? For the lab practical experiment, you will be given a 1.00-g sample of benzophenone contaminated with benzoic acid. Your task is to write your own protocol and complete as much of it as possible, with the objective being to obtain a pure sample of benzophenone in the highest percent recovery. Techniques to consider include the following:  Recrystallization  Extraction  Column chromatography  TLC/mp/bp  Distillation ...
