How can I find the fraction number in the gel filtration experiment?
in a biochemistry lab
Gel filtration chramatography is a technique which separates the proteein on the basis of their size. It is also known as gel permeation chromatography.
In this technique, stationary phase is made up of porous matrix and mobile phase is the buffer which flows between the matrix and beads. Beads have a defined range of pore size. So once protein get entered into the beads, it will restrict the movement of proteins. Higher the size of the protein faster would be the elution and vice versa because small protein get enetered into the beads and face more retention in the column.
So each and indiviual protein of different size will give a separate fraction, whihc can be collected manually.
