Kohler illumination is the principal technique for sample illumination in modern scientific light microscopy. It requires additional optical instruments. They are more expensive and may not be present in more basic light microscopes.

1. Lamp condenser focuses the image of the light source onto the frontal focal plane of the sub stage condenser.

2. The sub stage condenser focuses the image of the field diaphragm at the same plane as the focused specimen.

3. The filament image does not appear in the field of view and bright.

4. Illumination needs suitable adjustments of the condenser and of the field.
