PCR (polymerase chain reaction is a technique used in molecular biology to amplify a single copy or few copies of DNA segments into several thousands of DNA. The term PCR was first coined by Kary Mullis in 1985.

PCR technique completes in three steps: Denaturation, annealing, Extension.

In Denaturation of DNA results in unwinding and separation of the two strands at 92oC. In this step, the hydrogen bond which held together the two strands of DNA are broken down.
