In the presence of enzyme amyloglucosidase (AGS) starch is hydrolysed to D- glucose.
Again this glucose is phosphorylated into D-glucose-6-phosphate by ATP in the presence of hexokinase(HK) with formation of ADP. This reaction is done at pH 7.6
D-glucose-6-phosphate is oxidised by nicotin-amide adenine dinucleotide phosphate (NADP) to D-glucose-6-phosphate and formation of reduced NADPH
The amount of NADPH formed in above reaction is determined by means of its light absorbance is determined by means of its light absorbance at 334,340 or 365nm.
