In this blue white screeining, the plasmid contains gus gene (it also contians MCS or PLS), it produces b galactosidase enzyme which acts on X gal and turns the colony in to blue. When this plasmid inserted with foreign gene in gus MCS site, it leads to inactivation (it is reffered as insertional inactivation) of the product of gus gene, which fails to metabloize the X gal results the white color colinies.
