Cell membranes are asymmetric in nature with two phospholipid layers arranged in a specific
fashion. This asymmetry is first established in endoplasmic reticulum. Domains present on the
cytosolic surface of transport vesicles, golgi are found on the cytosolic surface of endoplasmic
reticulum. The luminal surface of endoplasmic reticulum is much like the extracellular space.
So based on all these facts, we can say that asymmetry of cellular membranes is established
initially in the endoplasmic reticulum.
