1. Culture based methods could not readily identify the large proportion of microbial population, whihc van be done by culture-idependent methods.

2. Most of the microbial population in unculturable thats why this method is not suitable.

3. This method has a lot discrepancy in its results in terms of its samplin, cultivation of microb's and transport.

4. It is very laborious and time consuming method.

Metagenomics: It is a study of genetic sample that has been recently recovered from the environment. It can also be termed as environmental genomics, ecogenomics etc.
