Answer #1

A dichotic mirror allows light of certain wavelength to pass while reflecting or blocking the light of other wavelengths. In a fluorescent microscope, the excitation and emission filters are present. The dichotic mirror reflects the light wavelength coming from excitation filter onto the specimen and it allows the en mission wavelength of light emitted from specimen to pass through it to the excitation filter. Thus dichotic mirror has the reflection of excitation wavelength and passage of emitted light functions. These are the functions of a dichotic mirror. An image is being attached. Hope it helps.

A total internal reflection fluorescence microscope uses a laser beam that undergoes total internal reflection within a glass microscope slide. The laser travels through a glass microscope slide and hits an interface with water. The index of refraction of water is 1.33 and the index of the glass is 1.52. What is the critical angle at which total internal reflection occurs within the microscope slide?
