UPDATED: Which of the following summarizes expression of the lac genes in this partical diploid: I+ P+ Oc Z- Y+ / I- P+ O+ Z+ Y- ?

The Minus (-) are on the first Z and the second Y. The Plus (+) are on the first I, P and Y and on the second P,O and Z. and the c is on the first O.

A) Both permease and beta-galactosidase are made both in the presence of and in the absence of lactose.

B) Permease is made both in the presence of and in the absence of lactose; beta-galactosidase is only made when lactose is present.

C) Both permease and beta-galactosidase are made only when lactose is present.

D) Beta-galactosidase is made both in the presence of and in the absence of lactose; permease is only made when lactose is present.

Answer #1

The answer is - B) permease is made both in presence of and in absence of lactose; beta galactosidase is only made when lactose is present.

In diploid genotype left side has Oc mutation so gene will give constitutive expression. So permease will form constitutively.

But the right side has I- so faulty repressor will produce so it cannot bind to the operator. But left side will produce active repressor that will bind to operator in the right side in absence of lactose.

So then the gene will not express.

1.    The lac operon in E. coli comprises three structural genes, lac Z, lacY, and lacA. It is regulated by both positive and negative mechanisms, involving binding of both a repressor protein and the catabolite activator protein (CAP). Transcription of the operon gives a polycistronic mRNA encoding the three enzymes. Under the following nutritional conditions, indicate whether or not the repressor and the CAP are bound by drawing them in on the appropriate regions of the operon. Indicate, as well,...

The lac operon in E. coli regulates the co-expression of three genes needed for the degradation of lactose (but absence of glucose) in the growth medium. These genes are not expressed when the bacteria has no lactose to feed on. How does the cell prevent expressing the lac operon in the absence of lactose? How does the presence of lactose activate the transcription of these three genes? (You do not need to explain the glucose level regulation.)

You isolated a mutant strain of E. coli in which the lac operon is constitutively expressed. To understand the nature of this defect, you created a partial diploid by introducing a plasmid containing a normal lacI gene (i.e. lacI+) and Z-. You then compared the mutant strain and the partial diploid with regard to their β-galactosidase activities in the presence and absence of lactose.
