Initially, labelled 14C acetyl CoA unergoes reaction with unlabelled oxaloacetate and this starts the TCA cycle. As the amount of 14C acetyl CoA is limited only one cycle occurs. All the intermediates in the cyle are labelled and finally oxaloacetate is also labelled. Carbon di oxide which emerges during the first turn does not have any labelling. This clearly means that carbon atoms of acetyl CoA do not correspond to the carbon of carbon di oxide, which is emerged during the first turn.
