The OCX-36 gene codes for the ovocalyxin-36 protein which has been shown to be secreted by the regions of the oviduct responsible for eggshell formation in chickens. You wish to study the genetics of egg formation in the, echidna, a mammal similiar to the platypus that makes you want to say, "Go home, Mother Nature, you are drunk." Specifically, you want to attempt to isolate the OCX-36 genehomologue from the echidna and study the about 50 kb upstream and about 50 kb downstream of the gene, looking for elements that may control its transcription. The echidna genome has not been sequenced, but you do have access to echidna tissue. Design and describe a scheme to find and isolate the echidna OCX-36 gene.

Answer #1

using sequence of OCX-36 gene different sets of primeres will be designed. If the gene is of 2kb than go for primers for 500bp region.

Or degenrative primers can also me made using different sequences of OCX-36 gene in different species.

Then using PCR gene will be emplified. desired gene fregment will be cloned using TA cloning and sequenced. If sequence matches with OCX-36, then full length gene will be emplified using 3'RACE AND 5'RACE.

Thus we have full lenght gene of OCX-36 gene homologue.
