The answer is - Urine sample.

Urine Sample may produce a mixed culture when urine gets contaminated by vaginal, skin or bowel organisms.

Therefore, care has to be taken while collecting the urine sample. The external genitalia has to be washed with soap and water and mid stream urine has to be collected in case of culture test.

While culture done from an agar slant of E.coli is a pure sample of E.coli and hence it does not produce a mixed culture.

Even stool sample too have less chances of getting contaminated and hence producing mixed culture.

Therefore, the answer is - Urine sample.
